Biomolecular Structure-Function and Systems Biology Characterization of Cell Stress Mechanisms

This project involves basic and applied research pertaining to defense in two areas: (1) structure/function studies of biomolecules and (2) systems biology and related studies (e.g., proteomics, metabolomics). In the first area, nuclear magnetic resonance and complementary techniques will be employed to probe structure, ligand binding, conformational changes, and intermolecular interactions of proteins, biological toxins, and enzymes of military importance. Complementary techniques may include molecular biology and biochemistry methods, as well as liquid chromatography tandem mass spectrometry. In the second area, systems biology approaches (proteomics, metabolomics, lipidomics) will be used to (1) follow protein expression and metabolic changes in cellular toxicity models of chemical or biological warfare agent exposure and (2) identify and characterize pathogens in various environments or under various growth conditions to understand mechanisms such as antibiotic resistance. Research may involve the use of bioinformatics and techniques mentioned above. The intent is to identify biomarkers of exposure to agent or to varying conditions and carry out pathway mapping to determine the biochemical pathways or mechanisms affected. This work supports the development of improved signaling strategies for detection and novel protection technologies.
